I need an extraction method for getting free fatty acids from whole blood. I don't want to strip the fatty acids that are membrane-attached and I only have 500 uL to work with.
Any suggestions?

Prep plasma/serum from the whole blood. Then extract into solvent. Then subject to SPE to separate the free/bound fatty acids.

Amino SPE cartridges are often used for serum phospholipid analysis. You could try the same method retaining the free fatty acid SPE eluate rather than the phospholipid eluate.
