Uracil is used for determination of the column void (dead) volume in reverse-phase chromatography, or in other words it is used to determine the dead time (the time for the unretained species to reach the detector).

Uracil is used for this purpose because it is unretained by the RP stationary phases

If I may add a comment to the answer, it has a good UV response and is easily detected by the commonly used UV detector.

There have been extensive discussions on determination of to (tm). One needs to be careful, uracil is retained by some columns under some conditions.
