Detector linearity calibration

Discussions about HPLC, CE, TLC, SFC, and other "liquid phase" separation techniques.

Based on recent information during an LC/GC webinar on AIQ, the presenter mentioned that you should have "global" type procedures for calibration so all instruments (HPLC) are being calibrated in a similar manner.

My question pertains to detector linearity. How does your company perform detector linearity, and does the procedure account for the different absorbance ranges of the different manufacturer models?

Does anyone simply use the ASTM procedure by running 5 standards, then calculating response factor? (Requirements being the rsd of the response factors must be less than 5%) Or does anyone simply calculate slope and intercept?

I'm just curious if there is an industry standard. Thanks.

Russell

Dionex uses 5 certified caffeine standards then calculates slope, intercept, r^2 and RSD of the linear fit. The specifications vary depending on the model, but for the UVD3400 they are r^2 > 0.9997 and RSD < 3.0%.

This test pre-requires that the detector noise and drift, the pump flow rate, and the injector repeatability all be within specs.
