What information we will get from Y-Intercept in method Validation

Quick answer:

Intercept above zero: interferences, blank peaks

Intercept below zero: adsorption of substance

There can be of course be other reasons for intercept not equal to zero (e.g not linear response). But if the respones is linear the explainations above are quite likely.

Regarding a reference, I think you can find it in a good LC book like Snyder-Kirklands "Introduction to modern liquid chromatography".

Also, you should compute the confidence interval around the intercept. If it includes zero, then you can consider the intercept to be zero for practical purposes.
