Hi All,

Just running a gradient method with the above mentioned column. Actual fact the method was developed and validated in my lab.

However, since validation and running the actual method on a large scale we seem to have had nothing but problems with poor chromatography.

We looked at everything possible, grade reagents, thourghly cleaning the system, extraction disks to filter the solutions. The last thing we are coming to is the column.

I have noticed that the physical design of the column has changed between batch 4 and subsequent batches. This seems to be when our problems have started.

Just wondered if anyone else had had any problems with this column. :?

Cheers
Anthony

Have you tried with other eqvivalent columns such as ACE C18 ?

GL Sciences (Japan) produces Inertsil packing and has sold it to numerous column vendors over the years. As a result, numeroud brands and hardware configurations have made it into labs. If you are having lot-to-lot variability issues, it may be time to try another vendor (or order directly from GL Sciences).

Also note - Varian<Ansys<Metachem - same columns, 3 different names due to corporate takeovers.
Thanks,
