plate count

Hi
I am a new user of the HPLC, and I have a new column for the HPLC. The column is an ACCQ.Tag amino acid analyis column. I am trying to do a plate count for this column following the instructions and I don't get a peek. Why??? I run a standard 0.05% Acenaphthene in acetonitrile and let it run 60/40 for 10 min the inject of 10 micro L. I get no peek.
Am I doing something wrong?

I don't mean to sound patronizing, but you said you were new to this, so I am checking all the possiblities.

Have you ever seen peaks on this system (with other columns, perhaps)?

Have you double checked to make sure your detector and data system are operating properly (set at the correct wavelength, sensitivity, etc)?

Are you sure your pump is working (mobile phase coming out of the waste line)?

Did you see a small baseline upset marking the dead time? If not, are you sure an injection actually ocurred?
