Column for highly aqueous mobile phase

Dear All

Can you recommend analytical column suitable to a highly aqueous mobile phase,less than 5% organic in HPLC analysis?

We've used a YMS-ODS-AQ 2.0 x 150 for two compounds that were highly hydrophilic. Organic was run at 4% with no problems.

You can try out Unison UK C18 - it's suitable for 100% aqueous
mobile phase, pH durable, and gets great column efficiency.

The Dionex Acclaim PolarAdvantage and PA2 are designed for 0-100% aqueous conditions. The Acclaim 300 C18 will work over the range 0-95% aqueous; this is a wide-pore type column. See http://www1.dionex.com/en-us/columns_ac ... p5669.html

One of our methods uses Supelco's Ascentis RP-Amide and we run a gradient starting at 1% organic.

Phenomenex has similar column technologies as well (Synergi Polar-RP, Synergi Fusion-RP).

Earlier in my career I had good luck with ODS-Aq as mentioned by an earlier post.

Bottom line - there should be a number of options available to you... Best of luck.

I believe the Discovery RP C16 Amide is one generation behind the above mentioned Ascentis Amide. This may mean a lower price compared to the newer Ascentis.

According to Supelco technical support you can run 100% aqueous (on the Discovery). The lowest organic we have used is 5% (MeOH) with very good results.

Another option would be to use C30 columns that have been shown not to have the dewetting problems the C18 columns have... Of course it always depends on your analytes...

There are several columns in the market for 100% aqueous mobile phase.

I have experience with Phenomenex Synergi Hydro-RP - that replaced Aquasil C18.
