Analysis of Dextrose and Mannitol

Dear Friends,

1) What is the best chromatographic condition for an injectable preparation containing a mixture of Sodium chloride,Dextrose, Adenine and Mannitol. The ingredient to be separated is Dextrose and Mannitol. I find that Sodium Chloride gives a peak along with Mannitol resulting in a broad combined peak with out the two separating.

2) on repeated injections, there is no reproducibility or consistency in the results obtained for Dextrose as well as the Mannitol peaks.

I use Prevail Carbohydrate ES 5ucolumn (Amino column), length 250mm, ID 4.6 mm, RID 10 A of Shimadzu.

Please give the a suitable chromatographic condition for the above product analysis.

You might be better off using one of the Ca++ form cation exchange columns (available from BioRad or Dionex, among others). Mobile phase is DI water. You could put a H+ form guard cartidge in line to take out the Na+.
