Calculating the Stock Solution Concentration for a Surrogate

Hello there,

I am running a small scale repeatability study on a project using LC-MS/MS and am trying to figure out what concentration I should add to my dry matrix for surrogate recovery. I would like to see my compound of interest at around 0.05 uM, my final resuspension volume is 100 uL and my injection volume is 5 uL.

You need to know which sensitivity you’re operating with. To do so, you’ll need to determine at least 3 concentrations of the compound you want to spike in your sample. Then it’s easy to inter- and even extra- polate to a desired amount.
