Can any one help me with the analysis of Vit B-12 in soft gelatin capsules with a claim of 2 mcg/ cap and in an oil based formula.

I feel HPLC condition is not a problem but the sample extraction is the real problem.
After you opened the cap and added your extraction solvent, cool the sample down and than filter it.

I require help in selecting suitable solvent for sample extraction.
Vit B-12 has aqueous solubility so can DMSO help to dissolve the oil in the cap ???
Yes the vitamin B12 will be soluble in the DMSO. You can use hexane to dissolve the oil, leaving the fatty acids/ triglycerides in this layer.
