Analytical recovery vit E determination HPLC-Fluoremetric

Hi,

The determination of vit E in Plasma needs extraction , so i prepare a standard solution of vit E and calibration curve using different concentration of vit E in ethanol ( 5-40 mg/l) , after that i prepar the same standard solution for the calibration curve but i replace the ethanol by plasma for the prerpartion, after extraction and analysis by HPLC-fluo .

The first calibration curve (vit E in ethanol) and the second calibration curve were ploted.

Can you plz help me , how recovery can be determinate ?

I would try using solvents for extraction that vitamin E is soluble in.

use Shodex ODP2HP column for better recovery from plasma sample.
any way,
compare the area between ethanol and plasma for the same concentration, and consider area in ethanol is 100%.
(area of vit E in Plasma X 100)/area of vit E in ethanol could give roughly the % recovery at each concentration.
