Analytical method for bortezomib isomers

Recently i was developed a analytical method on chiralpak-AD-H Column,peaks are well seperated.

these are stereo isomers,i was separated four isomers from each other in normal phase mode by using mobile phase N-Hexane,IPA,Etahnol and TFA in the ratio of 920:40:40:0.1v/v respectively,diluent is mobile phase.




Now problem raised with my lyophilized sample this is powder for injection,it is contain mannitol 35mg and my drug is 3.5mg,this cake is soluble in only water or DMSO.

water and DMSO is not miscible with my diluent because diluent contains 920 N-hexane.I was tried to use pure ethanol or IPA but peaks are broaden not separated.Peak shapes and

resolution is good when using mobile phase as diluent.
Try chloroform in which mannitol is insoluble and bortezomib is soluble. Filter solution, evaporate chloroform and dissolve the residue with your mobile phase
