ion-exchange chromatography

I need to purify a protein (4ml, ~ 5 mg total) by ion-exchange chromatography (DEAE-Sephadex). I got the protocol with buffers but don't the size of column I should use, how much beads I should use and how eluate volume I should collect. It there any website on these basics or anyone to help me?

We need more information about the information that you have. Please describe the protocol from which you are supposed to work!
