ion-exchange chromatography

I need to purify a protein (4ml, ~ 5 mg total) by ion-exchange chromatography (DEAE-Sephadex). I got the protocol with buffers but don't the size of column I should use, how much beads I should use and how eluate volume I should collect. It there any website on these basics or anyone to help me?
