Vitamine B1/B6 chromsystems question

Hello everybody,

I have a little bit of a problem with the Chromsystems B1/B6 HPLC kit and I thought you might help me.
I want to use the kit but it says that I need to switch the mobile phases at 2.5 min. Does anyone know if the switch is gradual or I just switch from 100% mobile phase A to 100% mobile phase B.

As for detection it is said that I need to switch at 4 min from one EX/EM setting to another. Can I just use an interval instead of switching between the two or it is mandatory to use the exact settings mentioned in the prospect.

I would recommend to follow the Chromsystems method since these methods are prouved to be very reproducible and precise. So please switch the mobile phase and the EX/EM setting as mentioned in the method. You can see the effect of switching EX/EM on the chromatogram. Although it isn't typical to switch mobile phases dramatically like this during a run, it looks like they are really accomplishing two types of analyses and capturing it in one chromatogram. The detector parameters are also switched accordingly to go with the change in mobile phase. If the manufacturer says its Ok to do this, I would go with it.

there are appliations for water soluble vitamins
i know you can find some at Nacalai, with their MS-II columns. they have 2 applications if memory serves me well
