I want to purify a product (amino group) which is analysed with a phosphate buffer ph:4,5 what is the nearest volatile buffer i could use to replace it for a preparative system.
Thanks

Use ammonium acetate at pH 4.5, and you have a real buffer (phosphate has no buffering capacity at pH 4.5). In order to prepare the buffer, use ammonium acetate, and then add acetic acid until the pH is at 4.5.

Will i get volatile residues in the collected solution which will be eliminated on rotavapor or lyophilisated ?
Thanks

It is reasonably volatile. Try it out! You will get faster results at elevated temperature, if your compound lets you do that.

Thank you very much, i will try soon to compare results for the separation...
