Need advice about separation of plant flavonoids.

I ve been working wit plant flavonoid extraction. I ve tried wit suitable solvents to separate but i wonder if i could use buffer system as eluents for separating flavonoids rather than using solvents..

Please your valuable advice needed..

Hi,

to get nice peak shapes with flavonoids, it is recommended to use encapped stationary phases and acidic buffers.

For example you can use an endcapped C18 column and formic acid at 0.1%, the flavonoids will elute approximately between 10-40% ACN.

If you are doing reversed-phase HPLC, then organic solvents such as methanol or acetonitrile (mixed with water) are required to control retention. In the case of ionizable compounds (weak acids or bases), an aqueous buffer is usually used in place of the water in order to minimize peak shape problems and help control selectivity.

can you tel us which ones are you trying to separate?

getting a good resolution is not only a question of solvents but also of column, some are tougher to do simply on a C-18 even of end-capped

Thank you for your replies.
I am relatively new to chromatography and i want to do flavoniod separation from the crude extract of sunflower root.
Since i am a fresher i ve to start with cellulose or alumina column.
I need suggestion that whether i could go with buffers in the place of solvent for eluting.

if you know which ones are there then you could more easily find a method.

all methods are different all use RP C-18 or alike columns and not cellulose or allumina, but again the right column will depend on the type of compounds
