Hello everybody,

i think i have some air stuck inside the column (C18 phenomenx Prodigy OSD3), I mobile phase all day, even with column point up, but i still have samll peak on the base line in the UV detector.

What can i use to get all the air out of the column?

Thanks a lot.

Matteo.

Could you please post a chromatogram of what you are seeing? It will help clarify the problem you are having. In the meantime, if you truly have an air bubble in the column, try flushing it with methanol.

Under what ever back pressure it is nearly impossible that you have an air bubble "in" the column. But if you have pumped air on the column this can cause a major problem. Please check your mobile phase bottles, all frits if they are glocked, check for leackage on your injection system, and than flush with ACN or Methanol
