problem about quantifying a natural lipid compound

I am trying to develop a method for quantification of a lipid compound in Human plasma.The compound is naturally present in human plasma.So I am confused even if I extract about 80 % how to calculate the results as it is already present in the plasma.

From your brief descripion, I can think of two approaches. 1) addition of a labeled standard and 1) if no labeled standard is available, standard additions.
