For digested colorectal cancer samples, isotope-labeled heavy peptides were spiked at 5 fmol/μg, with a total of 122 heavy peptides added. The injection amount was 1 μg peptides.
Results showed low signal intensity for isotope-labeled heavy peptides, averaging around 4.0e5, with only 19 light peptides successfully quantified. In contrast, the same sample analyzed on the Eclipse platform exhibited heavy peptide signal intensities around 3.0e6 and quantified 43 light peptides.
Our request: For the same sample on the 480 platform, we aim to surpass or match the quantitative results achieved on the Eclipse platform. We would like to consult whether anyone has encountered similar issues, potential solutions, and methods to further enhance signal intensities of both light and heavy peptides.
