How i can Separate the 3-hydroxy Acetophenone from 2-hydroxy Acetophenone and 4-hydroxy Acetophenone on reverse Phase HPLC with UV Detector.

I’m just curious: Do you get paid for developing methods of analyses? Or is this and other similar posts just a hobby of yours?

Actually i am working in the R&D Center of Analytical Development Lab and Also this is my Hobby to take the knowledge from all pesons which are avilable for discussion on Chromatography. what they are think about development. what are the basis for their Development. and then i find the litrature that why they are taking this kind of mobile phase, column and what are the other possibilities of buffer , column which we have . and also off-course if the method of analysis is available then we are going to validation to save the time which take the time for development .

Hi
Unless you absolutely have to use LC, you could try GC instead (1701 column). I used GC for purity of dihydroxy acetophenone, the selectivity incuded 2- and 4- mono hydroxy, unsure on the 3-hydroxy though.

C.K.S., probably most of us prefer it the other way around: That the lit. work is done first and then unclear parts discussed here. That way the discussion is of a higher grade, a lot of people will learn something in the process, and nobody has the feeling that he is doing someone elses work.

I think there is the problem of acute lypophylicity and same hydrophylicity of -OH group.
U sholud use C-18 or shield column / EPG phase. along with 20 to 25% methanol with THF (75%) against water
A should (Methanol +THF) : : 25: 75
B should water (may be 0.1% H3PO4) or Not
A : B : : 25 :75
wavelength -254 may work
Use this probanly elution order is 4,3 and then 2
