Analysis of zoledronic acid

Hi there,

Any of you knows how to quantify zoledronic acid (a bisphosphonate with an indol ring at the N-end)?. I have been trying ion-pair RP-LC, but I would like to know if is possible doing it with no ion-pair Chromatography.

IC would probably work if you have one available. May or may not give better sensitivity.

For zoledronic acid you can use UV with any buffer (pH 2-5)

Can you please help me with the chromatographic parameters you have used?
I am not sure where to start with mixed mode column.

Start with short column (Primesep SB, 50 mm in length):

1. Inject sample without column and measure peak area, after you inject compound with column peak area should match (for UV or ELSD) sum of all peaks you see in chromatogram

2. Your acid retains mostly by anion-exchange mechanism. In our method we used gradient but only to help elution of nonzwitter-ionic dronic acid. Presence of basic group helps elution of “dronicâ€

Do you think the tailing might reduce if I use a shorter column; 50mm instead of the 150mm I am using currently?
