Quantification of leucine in bacterial cells

Hello everybody!! :D

Do you know how can I make the quantification of leucine in bacterial cells? I want to remove bacterial cells from water samples, extract the leucine and make the quantification... Using HPLC how can I do this?

You need to analize Total Leucine or free?
This is importat to define for the sample treatment.
what kind of water you have? is drinking waters; sea water waste water?
Certainly you can use HPLC for that you can use precolumn or postcolum derivatation.
