I want to know how we can conclude that a peak of GC chromatogram is pure (contains only one substance)? What is the simply way to check / test the peak purity?

Other than using a mass spectrometer, (and even that is not 100%) another way is to use two dimensional chromatography using two columns with two vastly different selectivities with heartcut or other means to isolate a peak. That too may not be 100% certain.
You can approach 100% certainty with many analytes, but as a GENERAL question, there is no absolute way you can prove any peak is singular in composition.
best wishes,
