Hai,

Can any body suggest an HPLC method for Zoledronic acid which is a non chromophore compound.

Raman

This is highly hydrophilic biphosphonate compound.

I suggest that you try a ZIC®-HILIC column, using 80% acetonitrile/20% ammonium acetate buffer. Our experience with compounds like ATP are a promising indication for this application as well.

Your compound will retain similar to cidofovir (polar hydrophilic coompound with phosphate group). You can compare it to regular C18 column.

Being a hydrophilic bisphosphonate, retention in a reverse phase column will be very difficult. Also peaks may tail because of the Imidazole moiety in the compound. Also the compound will respond only in lower UV region. To solve these issues a quarternary ammonium ion pair (use phosphate or sulphate salts for UV cutoff) mobile phase using a 100% aqeous compatible reverse phase column will help you.
