Agilent prodedure for Amino Acid Analysis

Greetings All,

I was looking over the product manual from Agilent titled "Rapid, Accurate, Sensitive, and Reproducible HPLC Analysis of Amino Acids"

Has anyone had trouble with the autosampler mixing program at all? I'm thinking about getting another HPLC and the automated derivatization is extremely attractive for my purposes.

Has anyone tried this procedure on Wine?

May I suggest that you also look at the AccQ-Tag method from Waters? Off-line derivatization in seconds, 8 minute analysis time for the standard protein hydrolysate.
We managed to do this exact experiment with our WPS-3000 sampler. We did it with the pulled loop version, but it would also work with the split loop version. For derivatization we used OPA and FMOC and performed all reactions inside the vial. We also changed method conditions so that we separated all peaks as quickly as possible. Currently we have managed to separate of 20 amino acids within 7.3 min on an Acclaim 120 column (C18, 3.0 x 75 mm, 3 µm). If you want more details on this, just let me know.

I have analyzed wine (Pedrocelli '97 and Monthaven '98 ) using that method. I have a pdf poster of that wine analysis that I would be happy to email you.
