Hi, I'm new here and hoping I can find some helpful information. For the analysis of sample extracts for N-methyl-carbamates, I'm using an Agilent 1100 Series HPLC with Pickering post-column derivatization. It includes a quaternary pump, a fluorescence detector, and a C8 column, and I'm using both Acetonitrile/Water and Methanol/Water mobile phases. I have a few questions now and probably more later.

1) If anyone has used this type of system extensively, can you tell me what critical supplies I should always keep on hand? 2)What is the typical post-column pressure while in operation? (Mine is running around 220 psi, which seems high to me.) 3) If the pressure is too high, what is likely the cause? 4) What are the most critical maintenance items/procedures that will keep things running smoothly?

I'm completely new at this, so forgive me if these are tiresome questions for you. I have a hard time deciphering those manuals sometimes, and I think it might help to communicate this way.
