If I have a peak in TIC for morphine dissolved in methanol.
How can I quantitate it in mg%??

Simple.... with calibration

Prepare a sample of known concentration in the same matrix, e.g.10mg/mL in methanol. Run your unknown sample and the known sample and compare peak areas.

unknown peak area / known peak area x known concentration = unknown concentration in solution.

That's obviously a single point calibration, if you want more accuracy, run a calibration curve and create a linear regression equation.

If you want a weight% of a solid sample (e.g. drug) take the dilution factor into account when back-calculating.
