Dear friends
Mostly I found analysis of alendronate using ion-pairing reagent. I want to try for LC-MS/MS, and worried for using ion-pairing reagent. Again, I have to analyzed urine sample. Does anybody have idea for LC-MS/MS analysis for alendronate without using ion-pair reagent. If not exactly same, I will thankful for any comment on ion-pairing reagent using for LC-MS/MS.
Thanks in advance.

We have the technology which allows to analyze polar compound without ion-pairing reagents. Chehk the following links;

Here is the link to the chemistry behind the Primesep columns:

And also an application with similar compound -cidofovir (basic function and phosphoric acid residue):

Mobile phase is ACN/water/ Ammonium acetate (or ammonium formate, formic acid, TFA, etc.)

See other discussion;

"HPLC method for Zoledronic acid"

That is also a hydrophilic biphosphonate compound.

We have done similar compounds from plasma and urine by direct injection onto an Oasis mixed mode ion-exchange column directly connected to the MS. Check on our website or call Claude Mallet at Waters.

I am right now doing alendronate by LC-MS. Just use the USP method and instead of using a RI detector plug in your ms. WARNING: I advise you to use and orthogonal (or Z) source since there will be some deposits from the buffer
It is working fine with me. I have an Agilent MSD machine.

Ciao
