I believe that you may want to narrow your question to be eligible for a short answer. Or you may want to attend a seminar by LC Resources to learn.
Method development is a challenge topic, and there are many books written about it. Even if you have a book, you may not understand it all and still you may not be successful because your matrix is an impossible matrix!
