I use Agilent LC/MSD trap XCT,
ESI source, MRM mode.
There was no spectrum observed. I found only background signal at intensity about 20-80. I tuned with Agilent tuning mixture which have m/z 118, 223, 662,....... (I'm not sure the number).
Early problem checked and solved.
ESI chamber was cleaned.
Capillary was cleaned.
Spray from needle worked.
Vacuum pump (Rough and Turbo pump) worked
I found that current at Capillary and End cap offset were too high.
Capillary voltage set at -3500 V -----> Observed current was >2000 nA (Normal should be 20 - 200 nA)
End cap offset set at -500 V-------> Observed current was > 2000 nA
(Normal should be 20 - 200 nA)
Should these are the causes of problem of no spectrum found or another suggestions?
