incompatibility of ibuprofen

hello,friends.

someone knows if ibuprofen molecule has incompatibility with trichloro acetic acid.?

how can I test that question...

I ve some troubles with the quantification of ibuprofen in coated tablets,
I thinked that trichloroacetic acid used in diluting solution cause molecular degradation...

I saw some deformation ,but not always in my interest peak.

Hi,

From the structure of ibuprofen there is no obvious reason to think that it should be unstable in trichloroacetic acid. http://en.wikipedia.org/wiki/Ibuprofen

However, it won't be difficult to test it out. I would do the following:
First, make a standard solution of this compound, say 0.1 mg/mL;
Then, load two auto-sampler vials (1.5 mL) with equal volume (say, 0.95 mL) of above standard solution;
Then, add 0.05 mL of trichloroacetic acid to one vials, and add 0.05 mL of D.I. water to the other vial as the reference;
Finally, inject samples in both vials periodically using the same chromatographic conditions to monitor any change.

Did you have trouble with linearity result? If not, I would look into the sample preparation step. Can you provide more information on the sample prep. procedure, chromatographic conditions, and specifics of the "trouble"?

There should be no need to use TCA in the diluent which could cause a disturbance by being at a different pH to the mobile phase. I suggest using the mobile phase for your final dilution.

Since you are working with finished product, you may want to do a stress study on your API; excipient; and then
API + excipient. Sometimes excipients can degrade under certain conditions (e.g. light, acid, base, temp,ect.).
