Peak fronting - chiral method

We are transferring a chiral method for determiniing the S isomer content of an R isomer API. The column is a Chirex penicillamine and the mobile phase is 65% CuIIpentahydrate sln with 35% ACN. Std is prep'sd in the mobile phase. We are witnessing significant peak fronting. Changing inj volume does not help.

Maybe you’re dealing with column void volume/headspace.
Try to reverse the column and see if the fronting disappears.
