Bioanalysis of ester prodrug

I have recently been posed with the problem of developing an LC assay for an ester prodrug and it's active metabolise. Initial investigations have showed significant degradation of the prodrug post sampling which we obviously need to prevent as far as possible to gain accurate concentration results for both compounds.

I was wondering how other folk have approached this type of problem in the past. We have proposed chilled temperature during sampling and K oxalate/Na F anti-coagulant. The worst case scenario may be requiring to add an additive such as PMSF to the sample and in this case does anyone have experience in this being added to whole blood. I am concerned we may may have to resort to this but PMSF solubility and adding PMSF to blood in solvent are giving me headaches!
