Dear All Expert Chromtographers,

I am looking for an extraction method of propionic acid in bread. I would like to run it on a HPLC rather than GC due to their availability in my lab. Looking for any advises from you all.

Thanks and regards.

Dear Louise77

This is anextraction method, but analysis done by GC. Most of the literature ( try www.google.com) mention GC analysis due to nature of product and (non) detectability with UV

Gas chromatographic determination of propionic acid in bread and cake.Isshiki K, Tsumura S, Watanabe T.
A simple and accurate method is presented for gas-liquid chromatographic (GLC) determination of propionic acid in bakery products. Propionic acid is extracted with a mixture of dichloromethane and formic acid to which isobutyric acid has been added as an internal standard. The extract is injected directly into the GLC system. A glass column packed with 5% SP-1000 is used. The simplicity and convenience of the method make it suitable for routine determination.

Hopes this helps

There are possibilities, but they depend on the detection tools that are available to you. If you can do low-UV detection. I would suggest HILIC on silica (80% acetonitrile, 20% water on Atlantis HILIC silica). An alternative would be low UV detection with water with a small amount of phosphoric acid and an acid-stable water-compatible C18 (such as Atlantis T3). Both methods would also work with RI detection.

What about HPLC on an organic acid column with conductivity detection?
