plasma extraction method

Dear all,

Could anyone comment my plasma extraction method?

I used 80% NaOH 100 uL and then extract with ether 6 ml.
After centrifuged and transfered ether layer to other tube and evaporation, samples were reconstituted with mobile phase.

This extraction method was used for LCMSMS.

The problems are
1. Could NaOH contaminated to organic solvent layer?
Can I washed organic layer with water 6 ml?

2. How can I make the method more reproducible?

Can you justify the use of the NaOH ? I think you may be using for the drug to be unionised.
Why you can try with borate buffer? Check whether recovery with reproducibility is good.
