Hi
You prapare or choose sufficient quantity of sample with purity above 98.5 % .Analyse it as per specification.Prapare certificate of analysis. The assay value that you will be getting against reference standard is the value that you can use for the analysis of other routine samples.

If you want to prepare Reference standard then do Identification test by IR UV Mass NMR CHNS to conform its structure.Perform all the tests mentioned for that compound in the specification.Determine its potency by substracting Total known and Unknown impurities, Residual solvents/OVI , Water content/Loss on drying ,Solphated ash, Heavy metals from 100.
