Separation of Terbutanol and Adipic Acid

Dear Friends,

I need to find a HPLC column capable of resolving terbutanol and adipic acid. I know GC is the best way perhaps, but it is not an option for me.

Perhaps some of you have experimented with ion exclusion columns and know one that could resolve these two compounds. Biorad 87H retains adipic acid, but is not clear whether terbutanol would be separated.

Any suggestions?

That should be relatively straightforward with a RP column. Inject both, if they overlap, change the pH a bit. For standard UV, use a phosphate buffer and low UV detection. For other detectors, other buffers could be used.

If you go to Biorad's website and find the Guide to Aminex columns you will see a chromatogram of tert-butanol with a mobile phase of 0.01N sulfuric acid and RI detection. There are chromatograms of various organic acids. Adipic acid should elute before tert-butanol.

Terbutanol is totally soluble in water and can not be easily retained in RP phases. Adipic acid on the other hand is only slightly water soluble and can be easily retained even without pH control. The problem is to find a column that can retain and separate both without gradient conditions. Both have very weak Uv absorption and most likely it will be necessary to use RI.

I am still trying to locate a good ion exclusion column.

I would like to hear other suggestions.

I would look at the Silec columns for mixed mode seperation. If you run a low organic MP to retain the Terbutanol, you should be able to retain the acid with anion exchange.
