about Preparative Columns !

Hi all!

I'm going to purchase soon prep-columns, but I do not know much about them. Please give me some ideas. I would have information about the prep-columns which are kinds of typical or universal reversed phase (C18) and normal phase for the secondary metabolites purification. We are going to have each typical size of columns as small, middle and large sizes to fit the prep-HPLC system, so we have plan to purchase total 6 prep columns from 3 of reversed columns and 3 of normal phase columns.

Please give me some ideas or which specific columns I need.
I would appreciate any kinds of help.

What is the flow rate range and pressure range of your system, and how much sample do you need to load? Also, what is the sample solvent (metabolites sounds like water)? If the solvent is water, a silica column may not be a good idea.

Consider few factors when you decide on prep column:
1. Your compounds should retain and separate with prep friendly mobile phase (no ion-pairing reagent, volatile buffers which are easy to remove)
2. Column should provide you with maximum loadability
3. Stability of stationary phase (no contamination from hydrolysis of stationary phase)
4. Stability of silica bed in the column (no void formation/peak splitting after you use it for prep separation)
5. Stability of your compounds in the mobile phase during solvent removal (temperature, pH, etc.)
