there is drop in intensity when run in single transition

when the sample is run with single transition i.e of drug we r getting 10 lacs area of drug using instrument API MDS SCIEX 3200.But when run with any other transition along with the drug transition i.e in MRM mode ther is a drop in intensity.we r getting 2lacs area of drug.we have ramped all the gases,voltages then also we are not getting the intensity.
