I need a buffer composed of 4M guanidine hydrochloride/30% ethanediol (v/v)/0.01M NaOH for a protocol using Hydrophobic interaction chromatography, but this buffer has a very high density and it´s impossible to pass it through a 0.22 micrometre filter to use it in a FPLC system. Is there any secret I don´t get (these are my first attempts in the world of chromatography). there are many protocols using this buffer or even more concentrated ones. any suggestion??? :cry:
