I have read about ion pair chromatography. Can anybody make me know how it is used practically in HPLC and when it is required?

You can go to the FAQ for more information (the link is near the upper left of the screen, just under the Chromatography Forum logo).

The short answer is that ion pair chromatography allows both hydrophilic ions and more hydrophobic neutral compounds to be retained on a reversed-phase column under similar conditions. It is most useful when a mixture containing both neutral and ionized compounds must be analyzed.
