SPE sample preparation for Sudan dyes determination

Hello All,

Did anybody determines sudan dyes in chilli or paprika powders?
I would like to ask about cleaning sample with very complex matrix with SPE methode. I found ASTA methode 28 and some suggestion are discribed in it but I cannot to use ethyl ether for conditionig spe column. Is it possible to use any other solvents for preparations spe column for cleaning paprika powders for sudan dyes determinations?
Second problem is to drying sample - I don't have nitrogen for drying. Is it possible to vapour in any other way without degradating sample?

I used ethyl acetate and water for the extraction. I did not do any SPE cleanup. Instead, I used THF:MeOH mobile phase that simply elutes all the late eluting pigments at the end of the run. You can use clean, dry air to evaporate ethyl acetate because the Sudan dyes are quite stable. (Don't do that with ethyl ether; too much risk of ether peroxides.)

Till now I dissolve paprika or peppers in acetonitrill.
I'm using 150mm 4.6 mm 3um IMTAKT Cadenza column.
I will try to prepare samples in the way you discribed.
