Hi All,

It is observed that TFA(trifluroacetic acid) is commonly used in the prepartion of Mobile phase in drug discovery whereas in generic development it is usually avoided.

Can anyone point out any drawbacks of using TFA for method development in drug discovery process. Also does the use of TFA affect column performance.

I'm in generics & I use TFA a lot. There are better things to feed a MSD, so people who make extensive use of MSDs to help identify impurities may eshcew TFA as a rule.

I have found that TFA's effects on columns are generally very short lived and reversible - at least compared to TEA, sulfonates...
