Nucleotide Quantitation

I am doing a project on nucleotide quantitation. Basically using a method of perchloric acid extraction of 5'-mononucleotides from yeast extract followed by HPLC on a C18 rp-column. ref: Fish 1969. Looking for some ideas to improve the method to increase nucleotide recovery, time, etc. Tried pH change. Nucleotides are supposedly free, would an acid or enzyme hydrolysis help?

Any ideas greatly appreciated.
