HPLC-RID for sugar (50 ppm)

Hi,

I'm planning to analyze sugar (glucose, fructose) with the lowest concentration of 50 ppm. My instrument will be HPLC Agilent 1100 (with RID 1100 series also).

Is it possible to analyze sugar with RID for concentration of 50 ppm?
What is the possible column or mobile phase?

Hi,

Yes, it will be possible. You can find plenty of methods if you search for "HPLC-RI sugars".

An RID is too insensitive. The best concentration I could detect is 1000 ppm or 0.1% (30 years ago). Your better of to use a Biorad Aminex column (or equivalent) and 210 nm UV.

Check AOAC or ICUMSA (International Commission for Methods of Sugar Analysis) for an appropriate method based on your sample matrix and expected concentration.

We have also been looking into methods for Sugars and Dietary Fiber and they both use RID. I am wondering if LC/MS would be a good option or would there be problems ionizing the sugars?
