Recently I did the tuning and calibration of the API 3000 Triple Quadrupole Mass Spectrometer by using the standard positive PPG solution per the manual tuning tutorial of Analyst 1.6. Now Q1 can pass but Q3 would not able to pass for higher mass above 700 Da. When I do the Q3 scan, it is very strange that only less than 650 Da range show signal. It looks like that all the masses larger than 650 Da is cut for Q3. I returned the Q3 calibration and resolution values to factory settings and strictly follow up the Q3PosPPG.dam from "API instrument method" to do the tuning, still, mass above 700 Da could not pass. The I tried to optimize parameters such as ion source voltage, temperature, DP, FP, EP, RO2, CXP, IE3, etc., in a full scan from 50 to 2500 Da, still I could not see mass above 700 Da, no matter what kind of parameters I use, it looks like some settings make Q3 only detect the mass under 700. Please advise if this is the hardware problem. Thanks.
