I am setting up a Ultimate 3000 ACC-3000 autosampler, I have installed a 20uL loop and plan to do 1uL injections. I have Bottle D connected as wash solution.
I don't quite understand exactly what some of terms (the manual is vague), and if anyone can clarify the following it would greatly appreciated:
What is the loop wash factor?
Is the wash volume only from bottle D to wash inside and outside of the needle?
The flush volume is the amount of D pushed through the loop and valve to wash out the whole line before the next injection? Is there a good practice of how much volume compared with loop size to use?
Full settings:
Draw Speed 5 ul/s
Draw Delay: 3000 ms
Dispense speed: 20ul/s
Dispense Delay: 0ms
Dispense to waste speed: 50ul/s
Wash Volume: 150 ul
Wash Speed 50 ul/s
loop wash factor: 2.000
Inject Mode: Partial
Flush Volume 60 ul
If anyone has any comments on the other settings please feel free!
