Hi there. I'm taking a class on protein separation. My question is: if we glycosylate a peptide, would it decrease or increase the retention time in a reverse phase column?. Thanks for your time.

lsolines,

Carbohydrates are generally significantly more polar than peptides so a glycosylated peptide should also be more polar. Increased polarity will reduce retention on a reversed phase column.
