Can response of eluting peaks change with column phase?

I had installed DB-1301 column for analysis of cetostearyl alcohol assay test, it got failing results as stearyl alcohol which elutes later to cetyl alcohol would give percentage area response of 36% whereas limit was NLT 40%, then G2 phase was used (RTx-1) column which gave response of 50% for the same sample ...... :shock: :shock: ??? HOW???

If the Rtx-1 column is giving much sharper peaks then the ratio could be better. If the problem peak is broad on the 1301 phase then you could be losing some if it in an almost imperceptible tail, hiding some of the area of the compound in the baseline.

Thank You very much sir and yes as u said the peak eluted where over temp ramp starts and there was a elevation in baseline may be thats the reason ....
